Spermatocytology has become an emerging branch of male medicine. Through spermatocytology examination, not only can the spermatogenesis of the body be understood, but also the diagnosis of testicular spermatogenesis disorders and ductal diseases can be made, which plays an important role in both basic research and clinical applications of male medicine. Today, with the rapid development of male medicine, as research on the physiology, pathology, endocrinology, immunology, and male fertility regulation of the male reproductive system deepens, along with the improvement and advancement of light microscopy techniques, the application of electron microscopy in spermatocytology examination and the use of molecular pathology techniques have further enhanced our understanding and research on the morphological characteristics, ultrastructure, and related functions of immature germ cells, sperm, and non-germ cells. Current research has now reached the molecular level. In China, the incidence of infertility is as high as 15%, with half of the cases caused by male factors. Studies have shown that anatomical abnormalities of the male reproductive system, histological changes, dysfunction of reproductive physiology, immune dysfunction, and external influences are important reasons for the delayed development, morphological and quantitative changes, and functional abnormalities of germ cells. Therefore, observing the morphological structure of semen cells under physiological and pathological conditions at the level of light microscopic cytology and electron microscopic ultrastructure is of great significance for basic research in male medicine, eugenics, and the diagnosis and treatment of male infertility. In 1994, based on years of clinical work and experimental research, the author published the first domestic monograph on spermatocytology—Practical Color Atlas of Spermatocytology. After its publication, it was highly praised by experts both domestically and internationally. Ronald W. Lewi, professor at Georgia Institute of Technology and president of the International Society of Andrology, commented: "The atlas has clear and vivid illustrations, rich content, and fully reflects the author's profound expertise in spermatocytology." Stefan C. Müller, chairman of the Urology Department at Bonn University in Germany, wrote a letter to the editor praising the book as "an excellent summary of spermatocytology, an indispensable reference for male medicine researchers." Qian Shaozhen, editor-in-chief of Asian Journal of Andrology and president of the Asian Society of Andrology, and Zhang Zengbo, editor-in-chief of Journal of Urology and doctoral supervisor at Peking Union Medical College, among other renowned experts, also highly evaluated the book, stating that the images of sperm apoptosis reported in it were "the first in the world." The publication of this atlas was widely welcomed by researchers, educators, and clinicians engaged in male medicine research, teaching, and clinical work across the country, promoting the vigorous development of spermatocytology examination nationwide and partially replacing invasive testicular biopsies, thereby improving the clinical diagnostic level of male medicine in China. However, with the rapid advancement of modern science and technology, advanced methods such as electron microscopy, flow cytometry, molecular biology, and molecular immunology are increasingly penetrating into male medicine research and the diagnosis of male diseases. We felt a pressing need for a comprehensive and systematic atlas that reflects the morphological changes and ultrastructural characteristics of semen cells and related lesions. To this end, this book has been enriched, adjusted, and updated in content and chapters based on the original version. It includes additional images of ultrastructure in semen cells, content and images related to fluorescence in situ hybridization and immunohistochemistry techniques in semen analysis, and images of the occurrence, development, and evolution of spermatogenesis apoptosis under electron microscopy, as well as the formation and phagocytosis of apoptotic bodies by tissue cells. This newly compiled Atlas of Spermatocytology and Ultrastructure consists of 4 chapters and 19 sections. The first chapter introduces the occurrence, structure, and function of sperm cells, as well as normal and pathological spermatocytology and ultrastructure, and the classification of abnormal sperm and non-sperm cells. Chapters 2 and 3 respectively describe spermatocytology and electron microscopy examination techniques and their clinical significance, as well as cytological staining, electron microscopy preparation, and fluorescence in situ hybridization techniques. Chapter 4 is dedicated to image content. This book systematically introduces the application of advanced experimental methods and techniques such as spermatocytology classification, collection, staining analysis, electron microscopy, flow cytometry, molecular biology, and immunohistochemistry in semen cell analysis, along with histological images of common and prevalent male reproductive system diseases. The book contains 318 images, making it rich in illustrations, with novel and detailed content, strong practicality, and serving as an essential reference for clinical and experimental researchers in the fields of male medicine and reproductive medicine. In this book, all light microscopic images of sperm and non-sperm cells are uniformly magnified 1000 times, and magnification is no longer marked in the text, except for the images in the fifth section of Chapter 4.
