I don't know what microscope you're using. The one I'm working with requires the use of an oil immersion lens when magnified to 100x. That is, a drop of immersion oil is placed on the prepared slide, and the lens is then lowered onto it. It seems that this method provides clearer visibility. You can check the experimental manual for more information. However, after use, the lens must be cleaned with lens paper. Simply apply the semen directly to the slide—no staining is needed.
