Why is one dose of HPV more painful than the next?

Preventive vaccines primarily combat HPV infection by guiding an effective humoral immune response, specifically the production of neutralizing antibodies. As for the possibility that one shot may be less painful than multiple shots, it might be because each anesthetic injection is administered in the same location, and the area hasn't had enough time to recover yet!
