Proteins are the main and material basis of all life activities. With the continuous deepening of research in molecular biology, structural biology, genomics, and other fields, many scholars have shifted their research focus in the life sciences from genes to proteins, further promoting the development of proteomics and protein engineering. Protein research has become one of the most active areas of study in current biochemistry and molecular biology. To study the structure and function of proteins, it is first necessary to obtain highly pure, biologically active target proteins. However, proteins in tissues or cells generally exist in the form of complex mixtures, and each type of cell contains many proteins with different structures and functions, making protein separation and purification a meticulous and complex task. Although molecular biology methods can deduce the amino acid sequence and composition of the primary structure chain of proteins, many issues still require better explanation and refinement through the study of the spatial structure and expression patterns of proteins themselves. Additionally, using protein engineering to modify the structure of proteins also requires a fundamental understanding of their basic structure. Protein separation and purification is a field of great interest and long-term in-depth research for many biologists, chemists, and medical workers. Only after obtaining highly pure target proteins can their properties be studied, potentially leading to large-scale production and application to benefit humanity. Therefore, protein purification is not only very fundamental, important, and critical in basic theoretical research and revealing the essence of life phenomena but also essential for the industrial production of products such as protein engineering, genetic engineering, and cell engineering.

The book was written by Lu Jian, Zhou Nandi, Shi Feng, Cao Yu, Wang Dong, and Kong Weibao. These young teachers fully utilized their time for study and research abroad, combining their personal research experiences to gather and organize a wealth of materials. Li Yin, Zhao Haifeng, Li Wangjun, Cai Guolin, Feng Linglei, Liu Jun, and Wang Lu participated in the data organization and some computer text processing work. The entire book was compiled and finalized by Lu Jian. Due to the authors' limited expertise and the tight schedule, there may be many shortcomings in the book.
